Intestinal Organoid Built That Looks and Functions Like Real Tissue – Genetic Engineering & Biotechnology News

Organoids, which originate from stem cells, are a tool with great potential for modeling tissue and disease biology. The idea is to build miniature tissues and organs that accurately resemble and behave like their real counterparts. But there have been limitations to their development. A new study has taken organoids a step further by inducing intestinal stem cells to form tube-shaped epithelia with an accessible lumen and a similar spatial arrangement of crypt- and villus-like domains to that in vivo. These mini-intestines also retain key physiological hallmarks of the intestine and have a notable capacity to regenerate.

The work is published in Nature in the paper titled, Homeostatic mini-intestines through scaffold-guided organoid morphogenesis.

Organoids could complement animal testing by providing healthy or diseased human tissues, expediting the lengthy journey from lab to clinical trial. Beyond that, organoid technology may hold promise, in the long-term, to replace damaged tissues or even organs in the future. For example, by taking stem cells from a patient and growing them into a new liver, heart, kidney, or lung.

So far, established methods of making organoids come with considerable drawbacks: stem cells develop uncontrollably into circular and closed tissues that have a short lifespan, as well as non-physiological size and shape, all of which result in overall anatomical and/or physiological inconsistency with real-life organs.

Now, scientists from the group led by Matthias Ltolf, PhD, professor at EPFLs Institute of Bioengineering, have found a way to guide stem cells to form an intestinal organoid that looks and functions just like real tissue. The method exploits the ability of stem cells to grow and organize themselves along a tube-shaped scaffold that mimics the surface of the native tissue, placed inside a microfluidic chip.

The researchers used a laser to sculpt the gut-shaped scaffold within a hydrogel, a soft mix of crosslinked proteins found in the guts extracellular matrix supporting the cells in the native tissue. Aside from being the substrate on which the stem cells could grow, the hydrogel thus also provides the form or geometry that would build the final intestinal tissue.

Once seeded in the gut-like scaffold, within hours, the stem cells spread across the scaffold, forming a continuous layer of cells with its characteristic crypt structures and villus-like domains. Then came a surprising result: the scientists found that the stem cells arranged themselves in order to form a functional tiny gut.

It looks like the geometry of the hydrogel scaffold, with its crypt-shaped cavities, directly influences the behavior of the stem cells so that they are maintained in the cavities and differentiate in the areas outside, just like in the native tissue, said Ltolf. The stem cells didnt just adapt to the shape of the scaffold, they produced all the key differentiated cell types found in the real gut, with some rare and specialized cell types normally not found in organoids.

Intestinal tissues are known for the highest cell turnover rates in the body, resulting in a massive amount of shed dead cells accumulating in the lumen of the classical organoids that grow as closed spheres and require weekly breaking down into small fragments to maintain them in culture. The introduction of a microfluidic system allowed us to efficiently perfuse these mini-guts and establish a long-lived homeostatic organoid system in which cell birth and death are balanced, said Mike Nikolaev, a graduate student and the first author of the paper.

The researchers demonstrated that these miniature intestines share many functional features with their in vivo counterparts. For example, they can regenerate after massive tissue damage and they can be used to model inflammatory processes or host-microbe interactions in a way not previously possible with any other tissue model grown in the laboratory.

In addition, this approach is broadly applicable for the growth of miniature tissues from stem cells derived from other organs such as the lung, liver, or pancreas, and from biopsies of human patients. Our work, explained Ltolf, shows that tissue engineering can be used to control organoid development and build next-gen organoids with high physiological relevance, opening up exciting perspectives for disease modeling, drug discovery, diagnostics, and regenerative medicine.

Wealth: Who are the Richest Germans? – The Berlin Spectator

Germany has as many as 237 billionaires on the basis of the European currency Euro. An old acquaintance is still at the top of the list.

Germanys billionaires made their fortunes by selling hygiene paper, toys, beverages, industry cables, chicken, other food, packaging, campers, software, household appliances, real estate objects and sweets. Since Germany is the land of cars, it is no surprise that the owners of quite a few car parts suppliers made it on the list of the 1,000 richest Germans published by the Welt am Sonntag weekly.

Schwarz is Undisputed Winner

The very richest German national is Dieter Schwarz. It is not the first time he made it to the top of the list, with a fortune of 41.8 billion Euro (49.5 billion U.S. Dollars or 38.3 billion Pounds Sterling). His company, Schwarz-Gruppe, with an annual turnover of 113.3 billion Euro (134.1 billion Dollars or 103.8 billion Pounds), runs ths supermarket chain Lidl and Kaufland, a big box store chains.

About half Schwarz wealth is what the second-richest Germans, the Reimann family is worth. According to the ranking compiled by Welt am Sonntag, the owners of the businesses Coty and Jacobs Douwe Egberts are sitting on 21.45 billion Euro (25.4 billion Dollars or 19.7 billion Pounds).

Porsche and BMW Shareholders

Dieter Schwarz and the Reimann family were able to increase their wealth to some extent since 2019, in spite of or maybe because of the Corona pandemic. Third on the list is Wolfgang Porsche, one of the shareholders at Volkswagen and Porsche, with a fortune of 20 billion Euro flat (23.7 billion Dollars or 18.3 billion Pounds). In Porsches case, nothing much changed since last year.

Susanne Klatten remains Germanys richest woman, with a net worth of 15.1 billion Euro (17.9 billion Dollars or 13.4 billion Pounds). She is a shareholder at both BMW and Altana, a chemicals company. Her brother Stefan Quandt only hit the 9th place on the Welt am Sonntag list. He owns 13.6 billion Euro (16.1 billion Dollars or 12.5 billion Pounds).

Places Five to Ten

Dietmar Hopp, who is on the fifth place, made his fortune with software and bioengineering. Hasso Plattner, on the sixth place, is Hopps partner at the software company SAP. He also deals with art. The seventh and eighth places are being occupied by the families Heister and Albrecht who own the supermarket chain Aldi Sd.

The tenth place goes to businessman Heinz Hermann Thiele who manufactures brakes. At the same time, he is a shareholder of several companies, including Lufthansa. The people mentioned above are only ten out of a total of 237 billionaires in Germany.

Pale in Comparison

While there obviously is a lot of wealth in the Federal Republic of Germany, those on the list still look rather pale next to the richest Americans. Jeff Bezos fortune is about three times larger than Dieter Schwarz. Bill Gates, Mark Zuckerberg, Warren Buffet, Larry Ellison, Steve Ballmer, Elon Musk, Sergey Brin, Alice Walton and several other U.S. citizens are a lot richer than Germanys number one.

Research Roundup: Researchers test experimental COVID-19 antibodies treatment on patients – The Stanford Daily

Coronavirus antibodies treatment in phase II experimental trials

Stanford Medicine researchers are participating in a phase II multi-site clinical trial to test an antibody-drug designed to diminish early COVID-19 symptoms in patients with mild to moderate cases. Preliminary results are expected later this month.

The goal is to see if we can get sick people better faster, reducing both the length of their illness and how long they are shedding the virus, and therefore help prevent others from getting sick, Department of Emergency Medicine Chair Andra Blomkalns told Stanford Medicine News. I think this treatment shows great promise.

At Stanford Hospitals, the clinical site aims to enroll 20 to 40 patients who have tested positive for COVID-19. The experimental treatment involves using antibodies from patients who have recovered from COVID-19, then injecting these antibodies into COVID-19 positive patients.

Our hope is to effect treatment early in the course of disease, before it has time to progress further and potentially damage organs, Blomkalns said.

Microorganism yeast cells reprogrammed to produce plant-based drugs

Yeast cellular machinery can be genetically reprogrammed to change these microscopic organisms into factories that convert sugar and amino acids into plant-based drugs, a study published on Sept. 2 in Nature reports.

The drug shortages were seeing around the COVID-19 crisis drive home why we need new and more reliable ways to source these plant-based medicines, which take months to years to grow and come from a few countries, where climate change, natural disasters and geopolitical issues can disrupt supplies, bioengineering professor Christina Smolke told Stanford News.

The findings suggest that by making genetic modifications to the organelles found in yeast cells, the team can manipulate cellular processes to manufacture the desired chemical drug. In this case, researchers wanted to produce tropane alkaloid molecules for medicinal uses.

Plants are the worlds best chemists, Smolke said. We want to recapitulate their unique and useful chemistries in domesticated microbes to build complex molecules inspired by the natural world but tailored to better meet human needs.

Mosquito-borne disease likelihood increases as climate warms

In Sub-Saharan African, malaria rates are predicted to decrease, while other mosquito-borne diseases such as dengue fever, will increase significantly due to warming climates and urbanization, a study published on Sept. 9 in Lancet Planetary Health reports.

Climate change is going to rearrange the landscape of infectious disease, biology assistant professor Erin Mordecai told Stanford News. Chikungunya and dengue outbreaks like weve recently seen in East Africa are only becoming more likely across much of the continent. We need to be ready for this emerging threat.

The Aedes aegypti mosquito can transmit a wide variety of diseases, including Rift Valley Fever, yellow fever, Zika, chikungunya and dengue. The findings suggest that Aedes aegypti breeding grounds have expanded due to urbanization and warming climate. The mosquitos enjoy living in human-made water containers, like buckets or water storage tanks, and warm temperatures.

Its vital to focus on controlling mosquitoes that spread diseases like dengue because there are no medical treatments for these diseases, pediatrics professor Desiree LaBeaud told Stanford News. On top of that, a shift from malaria to dengue may overwhelm health systems because diseases introduced to new populations often lead to large outbreaks.
